Plate Location pin - A green pin that shows the well location used to auto adjust the probe

height for the main plate, reservoir, and/or strip wells. The default (and recommended)
location for the 96-well plate is well D6.

• The main plate depicts a 96-well microtiter plate. For information about the plates used

in assays, see

Strip Well depicts the strip well added to the off-plate reagent area when extra wells are

required. A strip well is equivalent to one column of a microtiter plate, and, like a
microtiter plate, its wells can be flat-bottomed, v-bottomed, etc.

Reservoir depicts the reservoir added to the off-plate reagent area when procedures

require larger amounts of a reagent such as alcohol or distilled water.

Move Probe Down - Moves the probe down in the specified well(s).

Plate Type - 96-well or 384-well.

Plate Name - Type a name for a plate for future use or select an existing plate from the list.

NOTE: If you save a plate without performing an auto adjust, the plate name

is saved, but the probe height adjustment is not. If you do this, you
cannot use the plate with this instrument. If your purpose is to save
this plate to use it when you create a protocol to export and send to
someone else, click Save. Otherwise, click Cancel and perform an
auto adjust.

Adjusting the Sample Probe Height

Adjust the sample probe height to ensure that the probe drops far enough into the well to
acquire sample.

NOTE: Ensure that there is no liquid in the wells or reservoirs before

adjusting the sample probe height.

NOTE: When you adjust and save the probe height settings for all three

areas under a plate name, all areas retain the adjustment.

WARNING:

Correct sample probe height is critical to successful sample
acquisition and calibration. Problems with the sample probe
height can lead to fluid leaks and inhibit sample acquisition.

1. On the Home page, click Probe and Heater under Daily Activities. The Probe &

Heater tab opens.

2. Select a plate from the Plate Type list.

3. If you are using a filter or mylar bottom plate, place a large (5.08) alignment disk into a

well. For other plates, no disk is required. Ensure that the well location is selected on the
plate image, (a green pin marks the location). To change the well location, click on the
desired well in the plate image.

4. Verify that the microtiter is not warped. Warped plates can lead to incorrect probe height

adjustment.

5. Eject the plate holder and add the desired plate.

6. Click Retract to retract the plate holder.

7. Type a name for the plate in the Plate Name box, or select a saved plate from the Plate

Name list.
