4.4.4 Setting up an Experiment
Open the lid of the amplifier.
Place the pMEA32S12-Lx chip inside the USB-MEA32-STIM4 amplifier. The pin layout is described
in chapter "Data Sheet” in the Appendix.
Close the lid.
Grounding the bath
The pMEA32-STIM4 is equipped with a big internal reference electrode, which will
be automatically connected to the systems ground when the amplifier is closed.
Connecting Perfusion Cannula and Temperature Controller
For setting up a complete perfusion system, a pump and a perfusion port for aspirating the liquid
from the culture chamber are required. Standard setups for MEA recording use a peristaltic pump
or syringe pump. The standard metal cannula with beveled tip for aspiration is fixed on top of the
USB-MEA32-STIM4 device beside the PH01.
Connecting Pressure Control
Connect the controlled vacuum pump CVP to the tube of the suction port. Please read CVP
manual, for setting up the controlled vacuum pump.
Alternatively: Connect a 1 ml syringe to the tube of the suction port.

Service and Maintenance
You should clean the contact pins of the amplifier carefully with a soft tissue and alcohol or
isopropanol from time to time, especially if you have problems with the noise level. For handling
and cleaning of the pMEA, please refer to the pMEA data sheet or to "MEA Cleaning Quick
Guide" on the MCS web site www.multichannelsystems.com.
Warning: Spilled liquid can damage or even completely destroy the electronics of the
USB-MEA32-STIM4 amplifier. Please be careful when setting up your perfusion system
and when starting the perfusion. Take care that the flow rates of the inlet and outlet
flow match so that flooding of the amplifier is efficiently prevented.
Please be very careful when handling the amplifier, or cleaning the device!
The pins can be damaged easily.
