The!figure!above!shows!the!front!of!the!NucleoCounter!SCC0400.!Two!operational!areas!of!
the!instrument!are!the!following:!

Sample!Pipette!
•
Run!button!and!an!LED!status!indicator!
The!sample!pipette!is!used!to!introduce!the!mixture!to!the!instrument.!
4.1 Instrument,Status,
The!LED!status!indicator!reflects!the!status!of!the!instrument.!When!the!instrument!is!not!
in!contact!with!the!PC!software,!then!the!LED!is!off.!When!it!is!on!it!reflects!instrument!
status!as!follows:!
•
Green!0!the!NucleoCounter!SCC0400!is!ready!for!operation.!
•
Red!0!the!NucleoCounter!SCC0400!is!busy.!
•
Flashing!Green!0!the!system!recommends!a!rinse!of!the!flow!system.!
•
Flashing!Red!0!the!system!recommends!a!clean!of!the!flow!system.!
4.2 Interactive,Controls,
The!Run!Button!on!the!left!hand!side!of!the!instrument!can!be!used!to!initiate!
measurement.!It!can!also!be!used!to!activate!other!operations!as!described!in!the!
following.!
4.2.1 Analyze-Sample-
Before!analysis!a!sample!must!be!prepared!and!the!sample!intake!immersed!fully!in!the!
sample!liquid.!Sample!analysis!is!activated!by!pressing!the!Run!Button!and!releasing!it!
immediately.!Upon!releasing!of!the!button!the!sample!is!drawn!into!the!flow!system!of!the!
instrument!and!is!finally!returned!to!the!waste!container.!!
4.2.2 Rinse-of-Flow-System-
The!primary!purpose!of!rinse!of!the!flow!system!is!to!flush!out!any!milk!containing!liquid!
from!the!sample!compartment!as!such!liquid!can!solidify!and!thus!compromise!the!
analytical!performance!of!the!system.!A!secondary!purpose!is!to!monitor!the!condition!of!
the!sample!compartment.!!
To!perform!rinse!of!the!flow!system!place!approximately!1mL!of!Reagent!C0400!in!a!sample!
container!and!immerse!the!sample!intake!of!the!instrument!in!the!reagent.!Press!the!Run!
Button!on!the!instrument!and!hold!it!pressed!until!it!starts!flashing!green,!then!release!the!
button.!!
4.2.3 Clean-of-Flow-System-
The!purpose!of!the!clean!of!the!flow!system!is!to!remove!material!from!the!sample!
compartment,!which!can!interfere!with!the!analysis.!
