Appendix B – Instrument Out-of-use Procedure

The fluid lines can become obstructed when the instrument is left unused for one week or longer. To avoid fluid line
obstruction, execute the following procedure BEFORE the instrument is left unused for one week or longer.

1. Flush and purge the lines.

These instructions are available with additional pictures in Service Procedure 208
located at

www.ankom.com.

1.1 With the instrument turned on, press the

Diagnostics button on the Touch Screen Display.

1.2 Press the

Line Charge button on the Touch Screen Display. The following screen will be displayed.

1.3 Fill the Enzyme Containers on the left side of the instrument with Distilled Water. The enzyme lines are first flushed

with Distilled Water to avoid any precipitation from occurring due to an interaction between the enzymes and the
EtOH.

1.4 Press the

Amylase button on the Touch Screen Display. This will draw water into the Amylase line. Do this a

second time to fully flush the line.

1.5 To flush the Protease and AMG lines, repeat step 1.4 using the

Protease and AMG buttons on the Touch Screen

Display.

1.6 Disconnect Enzyme, HCl, EtOH78, EtOH95, Water, and Buffer containers. Store or dispose of all fluids properly.

1.7 Obtain the TDF70 Line Flush Tubing Assembly shown below. Note the nine connection points on the Tubing

Assembly.

1.8 Attach the Line Flush Tubing Assembly to the instrument and to the 78% EtOH Container as shown above.

1.9 Fill the 78% EtOH Container with 220ml of 78% EtOH.

1.10 Press the

ALL button on the Touch Screen Display. This will draw 78% EtOH into all of the lines. Do this a second

time to fully flush the lines. This process will drain the 78% EtOH Container.

1.11 With the 78% EtOH Container empty, press the

ALL button on the Touch Screen Display to purge the fluid from the

lines by sucking air into them. Press the

ALL button a second time to fully clear the lines. There should now be

minimal fluid remaining in the lines. If a significant amount of fluid is observed, press the

ALL button again until

the lines are clear (some droplets will remain). During the last line charge you should see no fluid being pumped
out to waste.
