9. Steps 5 and 6 may need to be repeated several times in order to achieve
optimal protein purity.
If the sample needs to be clarified further, filter sample through a 0.45 µm filter.
10. Analyze the concentration of the target protein using a small amount of
supernatant.
Use SDS-PAGE to get a rough determination of the protein’s concentration.
This will help determine the appropriate size of the IMAC column needed.
11. Continue with purification protocols under denaturing conditions.
