Evaluate the data analysis performed by Experion software. Click on the Results tab
and view the RNA concentrations given for each sample in the Results table. See Test 1
for an example.

To perform a qualitative evaluation of quantitation reproducibility, overlay the
electropherograms of replicate samples. For example, in the virtual gel, select lane 3
and hold the Ctrl key while select lanes 6, 9 and 12. The electropherograms from wells
3, 6, 9 and 12 (500 ng/µl samples) overlap (Figure 2.3). Repeat for the other
concentrations. Tighter overlaps signify better reproducibility.

To perform a quantitative evaluation of reproducibility, calculate the coefficient of variation
(%CV), which is defined as the standard deviation/mean x 100. To do this, export the data
to a Microsoft Excel or other spreadsheet application either by entering the values
manually in Excel or by using the data export feature of Experion software as described
in Test 1. The data appear in the spreadsheet after sorting by peak number (Figure 2.4).
