7.2 The voltage activation curve of NaV 1.5

To determine the voltage activation relationship for NaV 1.5 a standard series of voltage steps
are applied at periodic intervals. Voltage steps of regular increments can be set in the voltage
command interface either from Wash/Test, A/D or Plate with acquisition. Currents recorded in
response to a series of voltage steps in an oocyte expressing the NaV 1.5 are illustrated in the
screenshot below. The voltage protocol is illustrated in the lower traces. Automatic detection
of the peak inward current can be done using Min-Max setting with the cyan and blue crosshair
positioned as on the figure.

Voltage steps evoke inward currents in NaV 1.5:

Inward currents are analyzed using the “calculator mode” and selecting the appropriate icon
corresponding to the voltage step protocol. Display of the results is obtained by selecting the
icon indicating the typical IV curve of a sodium channel.

The representation of the loading plate with oocytes recorded for the given experiment setup
is represented and each single IV curve is shown as a “mini” graph as the following screenshot.
In this experiment a single oocyte was probed and the desired IV can be drawn by selecting the
“mini” graph represented on the left of the figure above.
