Use the number keys to input the length of the oligo and press Enter. You can
enter a length of up to 99 nucleotides. After you press Enter, SmartSpec Plus
will calculate and display the extinction coefficient and molecular weight.

The molar extinction coefficient is displayed with units of liter mole

Press the right arrow to continue the assay. SmartSpec Plus will now be ready
to measure the absorbance of your oligonucleotide and to convert those
absorbance data into concentrations of µg/ml and pmol/µl.

b. Composition.

Use the number keys to input the number of each type of nucleotide. Use the
arrow keys to move from field to field. Press Enter when all four have been
specified. SmartSpec Plus will calculate and display the extinction coefficient
and the molecular weight.

Press the right arrow to continue the assay. SmartSpec Plus will now be ready
to measure the absorbance of your oligonucleotide and to convert those
absorbance data into concentrations of µg/ml and pmol/µl.

c. Sequence

SmartSpec Plus will automatically shift to the Alpha mode on the keypad so
that you enter an A by pressing the 3 key, a C by pressing the 6 key, a G by pressing
the 9 key and a T or U by pressing the exp key. You may enter a sequence of up
to 60 nucleotides. As you enter sequence, it will scroll to the left. You can move
around in the sequence with the arrow keys. Put the cursor under a nucleotide
and press Cancel to delete it. Put the cursor under a nucleotide in front of which
you want to make an insertion, then type the base to be inserted. Press Enter
when the sequence has been completely specified. SmartSpec Plus will calculate
and display the extinction coefficient and molecular weight of the oligo.
