Create Qualitative Assay Protocol

You define each Qualitative protocol using a single kit. The kit must contain one standard.
The standard is assigned a “Quali” value for each test. Controls are optional, but are
recommended for judging the acceptability of batch results.

1. Open the Protocols page, then open the Protocols tab. Click Create New Protocol.

The Settings tab opens.

2. In the Name box, type the name of the protocol.
3. In the Version box, type the version of the protocol.
4. In the Manufacturer box, type the manufacturer information for the protocol.
5. Define settings in the Acquisition Settings section. For information about each of

these settings, see “Acquisition Settings” on page 25.

6. Define settings in the Analysis Settings section, selecting Qualitative as the analysis

type. For information about each of these settings, see “Acquisition Settings” on
page 25. The
protocol for a qualitative assay contains a single standard.

7. Click Next. The Analytes tab opens. For information about the settings on this tab, see

“Analytes Tab” on page 59.

8. Click the desired analytes (bead ID) in the numbered grid. Information about the

analyte displays in a list on the right side of the grid.

9. To change the Default Analysis, click Change. In the Analysis Settings dialog box,

click the analysis method from the Analysis Method list. Click OK to change the
default analysis to a single analyte. Click Apply to All Analytes to apply the selection
to all analytes. To edit an analysis setting, see “Edit Qualitative Analysis Settings” on
page 120. Skip this
step if you do not want to change the default analysis or if you
chose No Analysis on the Settings tab.

10.In the Units box, type the desired unit of measurement.
11.In the Count box, type the total desired bead count for each analyte. Click Apply All.
12.Click Next. The Plate Layout tab opens.

To add well commands, highlight the appropriate wells and mark them as unknown,
standard, control, background, or wash. You can also delete commands that you’ve

added, and change the starting location on the plate. If you wish to run in replicate,

change the Replicate Count to the appropriate number and the grouping to your

preferred grouping method.

To add maintenance commands, choose a command from the list. Highlight the well
that you want to apply it to, and then choose Before Well Command or After Well

Command. If you are working with more than one plate, choose Add & Change Plate.

Here, you can add a plate, change the order of the plates, and scroll through all plates.

As you add commands to your plate, they appear in the command sequence chart.
Here you can give each well an ID. You can also import an ID list and move commands

for unknowns up and down in the command sequence chart.

13.Click Save.

Edit Qualitative Analysis Settings

1. Open the Protocols page, then open the Protocols tab. Select a protocol and click

Edit.

2. On the Analytes tab, click an analyte’s Analysis field on the right side of the window to

open the Analysis Settings dialog box.

3. From the Method list, click Luminex Qualitative or No Analysis.
4. From the Formulas list, click Lum Qual or Adv Qual.
