Leica CM1860/CM1860UV – Cryostat

7. Daily Use of the Instrument

7.1 Specimen freezing

• Select the sectioning temperature (chamber temperature) according to

the tissue type to be sectioned (

see "7.4 Temperature selection chart"

).

7.1.1 Quick freeze shelf

The cryochamber is equipped with a quick freeze shelf (5, Fig. 24) on which

up to 10 specimen stages with specimens can be attached.

The temperature of the quick freeze shelf is always lower than the cryo-

chamber temperature.

• Cut the specimen to size.

• If necessary, enable the Peltier element (4) - it

takes approx. 60 seconds until the maximum

cooling capacity is available.

• Apply a sufficient amount of cryocompound

to a specimen disc to orient and mount the

specimen.

• Place the specimen disc in one of the holes of

the quick freeze shelf and freeze the specimen

at a low temperature.

• Once the specimen is frozen, insert the speci-

men disc (

Fig. 25

) with the specimen into the

specimen head (

Fig. 25

) and start sectioning.
