Creating Calibration Libraries

16.1 Introduction To Quantitative Analysis

Quantitative analysis involves building a method, creating a calibration library of
target compounds, and associating target compound responses with concentration
results.

A CMS5000 method can be developed to collect and quantify sample data. When
developing methods, certain criteria must be met. For example, the method must
produce acceptable retention times, sufficient compound separation and detect
compounds at the desired sensitivity. These parameters will vary depending upon
the application. Refer to

for additional information on

building a method.

The Calibrate function sets or defines the calibration library which is the key
component in the quantitative method. The library contains the analyte name,
analyte area, the retention time, which is the time it takes the analyte to elute from
the column, and the response factor used to calculate the concentration of the
analyte. In water methods, the water beta factor (coefficient) and water
temperature are also taken into consideration when calculating the concentration.

The process of building a quantitative analysis method:

Prepare multiple standards at various concentrations.

WARNING

Wear appropriate Personal Protection Equipment (PPE)
as advised by the MSDS of the standard(s) being used.

Analyze the prepared standards with the CMS5000.

Identify the compounds by retention time in the analyzed standards with the
calibration software.

Build a target compound library.

Calibrate the library.

Verify the accuracy of the quantitation with known standards.

Analyze unknown concentrations to determine the presence and concentration
of target compounds.
