5.13.1 Procedure (continued)

• In contrast to "wet" coverslipping, higher ap-

plication quantities of mountant are necessary

for "dry" coverslipping, i.e. the parameters VOL

and/or TYP must be increased as needed.

• If this is no longer possible, a dispenser needle

with larger diameter can also be used.

4 Preparing the coverslipping machine

• Be mindful of air-tightness when insert-

ing the coverslip mountant bottle (

refer to

Chapter 4.11

).

• Insert a suitable dispenser needle (

refer to

Chapter 4.6

).

• Check the dispenser needle height and adjust

as needed (

refer to Chapter 4.7

).

• Carry out the priming cycle (

refer to

Chapter 5.3

).

• Fill the dispenser needle cleaner, being mindful

of the correct seating (

refer to Chapter 4.8

).

• Fill the loading bath with a suitable solvent

(

refer to Chapter 4.11

).

• Fill the cover slip magazine (

refer to

Chapter 4.10

).

5. Important information regarding test run

procedure for determining the optimum cov-

erslipping parameters

6. Evaluation of coverslipping quality
The respective coverslipping quality of the test

runs should be checked at various points in time

for adhesive and air bubbles:

Directly after coverslipping operation.

1 - 3 hours after coverslipping.

24 - 48 hours after coverslipping.

Should air pockets appear between the speci-

men slide and cover slip after these checks, the

settings must be corrected.
If air pockets still occur despite suitable chang-

es,

Chapter 8.2

provides additional directions.

• The initial coverslipping trials

should be carried out with empty

specimen slides, i.e. without a spec-

imen.

• If coverslipping quality is good, sub-

sequently carry out a test series us-

ing an applied specimen.

• Since factors such as section thick-

ness and specimen composition and

type can affect the coverslipping

quality, it may be necessary to make

minor corrections to the parameter

settings.

Ex. VOL: Increase or reduction of ad-

justment by ± 1

and/or TYP: Increase or reduction of

adjustment by ± 1
